Now if they could not get the rights to Peter Pan and so did not get Hook, I think Gremma would have been endgame, as that was likely their Plan B. Graham and Hook are rather similar. I think they started with Graham, got the rights to Hook and so scrapped Graham (poor boy).
I hate to tell you, but you have a lot of things that are factually incorrect here. Graham was always going to be killed off, Jamie (the actor) came into the role knowing he was going to be killed off in the first season. The rights to Peter Pan were not confirmed until right before the writers had their planning week for Season 2. Also, Colin was not originally a regular, he is billed as "guest" until about halfway through Season 2.
To answer the original question of this thread, Emma and Regina would have never been done, because the characters are both straight. While I get that they could have changed this, since the characters are fictional, I think it would have been a bit of a stretch, as (despite what some people saw) the characters never showed anything that would imply they were not straight. Even Red being bi felt a bit forced, and she had way more wiggle room because we didn't see her as much.

Billed as guest but they had promoted him to regular before his first episode. A&E have also said that they had to wait and see if Jen and Colin had the right kind of chemistry.

Talking in-universe (as in, if the characters were real life people) I don't think Emma would have got with Regina even if Hook wasn't an option.

Besides being Emma's first love, he was both Henry's father and Rumple's son/main motivation, heavily connecting him to some of the series biggest plot threads in the earlier seasons. Since his re-introduction in 2x14 and throughout the remainder of Season 2 and 3A, the writers seemed to have been building up to the two of them eventually getting back together. His death felt random and rushed, and a cheap means of getting him out of the way so that Emma could be with Hook.
Don't think so. I don't think Neal was ever written to be Emma's love interest. He was written as her tragic backstory in my humble opinion.
Colin broke his leg in late s2, so Hook had to be written out and was replaced by Neal, which gave him a bigger role than was orignally intended

I doubt it. I don't think Swanqueen was ever even on the table seriously, and it seems to be totally fan-based. Without Hook, Colin could have still been on the show as another character. Maybe he would have been the Wizard of Oz instead, or another character we eventually saw.
If they wanted to go for Emma having a girlfriend, if she hadn't been added so late, Lily would have been more likely (though I saw zero sparks chemistry between the actresses.) Lily's backstory and similarity to Emma's, all while being the daughter of Maleficent would have made sense, but adding her in so late would have felt a bit off.
I was never a fan of Swanqueen as a romance, but I love them as best friends and I think even without romance them raising Henry together as a family in a single household could have been a thing.
However, Emma was destined to be given a Disney princess ending while Regina's happiness was elsewhere and her romance left open.
I still think Colin as another character or a similar character played by someone else would have been added in to take Hook's place. The timing was better, it set up the drama with Neal and pushed the long game the writers were looking for.
